About This Tool

This calculator simulates the equilibrium between Protein A, Protein B, and an inhibitor that binds A in a competitive binding system. It computes the fraction of bound and free species based on user-defined dissociation constants and concentrations. The results include numerical values, visual pie charts, and a curve showing the effect of increasing or decreasing inhibitor concentration. Please contact Stephane Betzi for suggestions.

  • Input Parameters: Enter the dissociation constants (KD) and initial concentrations for Protein A, Protein B, and the Inhibitor.
  • Goal: The system solves equilibrium equations using a 2D Newton-Raphson numerical solver, estimating bound complex, remaining complex in the presence of an inhnbitor, as well as free species ratios.
  • Calculation: Check the "Math" button at the bottom left of the screen for calculation details.
  • Visualization: Graphical outputs help interpret binding competition and inhibitor efficiency relative to starting [Protein A], [Protein B], or [Inhibitor] concentrations ([A]0, [B]0, or [I]0).

DISCLAIMER: The javascript solver may struggle with extreme values. Choose an appropriate unit, check for aberrant results and for non-sigmoidal curves in the bottom graph (convergence failures).
